Choosing the right antibody is critical to the success of your immunoassays. In the course of assay development, choosing the right antibodies with high affinity and low off rate at the start of assay development is critical. Biacore analysis examines the characteristics of your antibody—antigen interaction. It uses Surface Plasmon Resonance (SPR) technology for the label-free, real time measurement of association (on) & dissociation (off) rates (ka & kd) and binding affinity constant (KD) of your antibody antigen interaction. Incorporating Biacore scouting assays early in the antibody screening process allows you to rank your monoclonal antibodies based on their binding kinetics and help you select the right antibodies for your assay. A relative epitope scouting assay with Biacore SPR technology can easily identify antibodies that bind to distinct separate epitopes on your antigen. Biacore assay helps you establish a panel of monoclonal antibodies with known binding kinetics to different epitopes of the antigen. For sandwich immunoassay development, it is easy to identify a pair of antibodies that can be used as capture and detection antibodies. A full binding kinetic analysis can then be performed on your final assay candidate antibodies.
Biacore assay is not limited to the characterization of antibody antigen interactions. Biacore assay can be also used for determination of label-free binding and a real time analysis of the strength of any molecular interactions provided that the molecular mass can be detected by the instrument (molecular mass above 1,200). The utility of Biacore assay includes, but is not limited to:

Antibody Scouting Analysis (ranking by affinity)
Biacore scouting analysis of antibodies are carried out using Biacore 3000 in order to rank the selected mAbs according to its KD calculations. Certificate grade CM5 chip are coated with capture antibody (goat anti-mouse IgG Fc for mouse mAbs, goat anti-human IgG Fc for chimeric, humanized or fully human antibody). The test mAb is then captured on the chip and analyte (antigen) at a saturating concentration will be passed through to measure rough ka and kd in real time. The KD will be determined from the ka and kd values obtained. Based on the kinetic parameters, test antibodies can be ranked according to their binding affinity and dissociation rate (off-rate or kd).

Full Binding Kinetic Analysis
Biacore full binding kinetic analysis of antibodies are carried out using Biacore 3000. Detailed binding kinetic parameters (association rate, ka, dissociation rate, kd, and affinity constant, KD) can be determined by full kinetic analysis. Certificate grade CM5 chips are coated with capture antibody (goat anti-mouse IgG Fc for mouse mABs, goat anti-human IgG Fc for chimeric, humananized or fully human antibody). The test mAb is then captured on the chip and the binding kinetics will be measured at up to 6 different analyte (antigen) concentrations (from 0 to saturating concentration). Binding kinetic parameters (ka, kd, & KD) at each analyte concentration will be measured and a χ2 analysis will be performed to assess the accuracy of the data. In our experience, high affinity antibodies with low off-rate exhibit good utility in immunoassays. The following is a typical Biacore analyses:

Relative Epitope Mapping
Biacore epitope scouting analysis of antibodies are carried out on Biacore™ 3000 for Epitope Mapping in order to identify mAbs that recognize distinct separate epitopes on a single antigen. Certificate grade CM5 chips are coated with capture antibody (goat anti-mouse IgG Fc for mouse mAbs, goat anti-human IgG Fc for chimeric, humanized or fully human antibody). In the first step, mAb is captured on the chip and analyte (antigen) will be passed through to allow binding. A second mAb will be applied to the Ab-Ag complex to examine binding. If the second antibody binds to a separate epitope, we expect to observe additional binding on the sensorgram. The following is a typical relative epitope scouting sensorgrams:
Alternatively, scouting assays can be carried out by capturing the antigen on the chip and sequentially passing the antibodies over the antigen to observe antibody binding. By measuring antibody binding using combinations of test antibodies, antibodies binding to different epitopes and to the same epitope are identified.
